I have many, but three that immediately come to mind are...

The scene when Eva Amurri finds Macaulay Culkin at the outdoor cafe in Saved!.

The scene when Gwyneth Paltrow gets off the bus and is met by Luke Wilson in The Royal Tenenbaums.

The scene at the graduation when Natasha Lyonne does her cheer for Clea DuVall in But I'm a Cheerleader.

I second "Persuasion". "I tried to forget you. I thought I had." **melt**

I also like the scene in "Sense and Sensibility" between Edward (Hugh Grant) and Elinor (Emma Thompson) when Edward says, "...my heart is--and always will be--yours." **sigh**
